题 目:Joint Analysis of Multivariate Longitudinal Data and Interval-Censored Event Time Data with Application to Huntington’s Disease Progression
摘 要:Huntington's disease is an autosomal dominant neurodegenerative disorder, characterized by motor dysfunction, psychiatric disturbances, and cognitive decline. The onset of Huntington's disease is diagnosed by severe motor impairment, which can be predicted by cognitive decline and may also worsen cognitive impairment. However, clinical data are often collected at discrete times, and therefore, disease onset is subject to interval censoring. We develop a joint model of multivariate longitudinal biomarkers with a change point anchored at an interval-censored event time. Our model allows us to simultaneously study the effect of longitudinal biomarkers on the event time and the changes in trajectories of the longitudinal biomarkers post the event. We conduct a comprehensive simulation study to demonstrate the satisfactory finite-sample performance of the proposed method for making inferences. We applied the method to the PREDICT-HD data from a multisite observational cohort study of prodromal Huntington’s disease individuals to ascertain how cognitive impairment and motor dysfunction interact during disease progression.
个人简介:Dr. Ying Zhang is a professor and Chair of the Department of Biostatistics at the University of Nebraska Medical Center. He earned both his B.S. and M.S. degrees in Computational Mathematics from Fudan University and his Ph.D. in Statistics from the University of Washington. Dr. Zhang is an elected Fellow of the American Statistical Association in recognition of his outstanding leadership in statistical graduate education and his foundational contributions to statistical methodology, particularly in the areas of panel count data and interval-censored data. His research has been published in leading statistical journals, including the Annals of Statistics, Journal of the American Statistical Association (JASA), Biometrika, and Biometrics. In addition to his methodological research, Dr. Zhang has made substantial contributions to numerous NIH-funded biomedical research projects through his statistical expertise. His methodological work in Huntington’s disease research has had a significant impact on the field. Throughout his career, he has also provided leadership and service to several professional statistical organizations, most notably the International Chinese Statistical Association (ICSA).
